The Roche 454 Genome Sequencer is a next generation sequencing platform. The complete system comes with an instrument, a high performance computer, a software package for data acquisition, processing, assembling, a bead counter and a waste bottle. It is a bench top instrument. The 454 GS Junior combines emulsion polymerase chain reaction with pyrosequencing chemistry. As the DNA chain is elongated by nucleotide incorporation light is emitted. In pyrosequencing there is no need for gels dideoxynucleotide triphosphate and fluorescent dyes. It can generate thousands of reads in a single run, at a much lower cost, higher accuracy, efficiency and sensitivity. In addition, the 454 GS can generate average read lengths of about 400 base pairs of high quality (99%) and up to 600 bases pairs.
